Mu - just 19 - clocked one minute, 55.21 seconds, a new American record.
Briton Keely Hodgkinson (1:55.88) - also just 19 - claimed silver.
Mu's American teammate Raevyn Rogers (1:56.81) came from nowhere to clinch the bronze on the finish line and give the Americans a double celebration at the Olympic Stadium in Tokyo.
It marked the first time an American woman had won the 800m Olympic title since Madeline Manning in the 1968 Mexico City Olympics.
The absence of South Africa's double champion Caster Semenya - ruled ineligible due to heightened testosterone levels - opened up the field to a new winner at Tokyo.
And Mu took full advantage.
She dominated the competition from the outset at Tokyo, winning both her heat and her semi-final.
She was never seriously challenged in the final as she strode away down the home straight to claim gold at her first Olympics.
"Coming here, I came with a medal mentality," Mu told reporters at Tokyo.
"I came here wanting a medal."
