Officer in charge of traffic operations Sergeant Rod Anderson said it appeared the motorcyclist died instantly.
"This has been one of the worst collisions we have seen in the last few years," he said.
"It was about 4.50pm when a motorcycle and a heavy vehicle were involved in a fatal collision and the motorcyclist was killed."
The crash happened near the Angle Crossing Road intersection.
It is the ninth death on a Canberra road this year.
The motorcyclist is a man and he is believed to be from Canberra, but police declined to reveal any further details as they were yet to notify the next of kin.
Sergeant Anderson said the truck driver was in a "devastated state". While he had not been injured in the crash, he "has been badly shaken". He is receiving support and counselling.
He said the motorbike and truck had been heading in opposite directions on the highway, and had collided at high speed. However, he said it was not clear if speed had been a factor in the crash.
The collision and investigation reconstruction team was examining the scene to determine how and why the crash happened. Officers were also interviewing several witnesses who saw the crash.
